dc.description.abstract我們於逆流層析儀中使用反微胞微乳液作為靜相,逆流層析管柱容積28 mL,使用含有100 mM sodium bis(2-ethylhexyl) sulfosuccinate (AOT)的n-hexane作為靜相,動相是酸鹼值以及離子強度梯度變化的緩衝溶液,動相流速1mL/min。在800 rpm轉速下,靜相滯留於管柱中的量為12 mL,分離1 mL含有肌紅蛋白、細胞色素C、溶菌酶各2 mg的蛋白質混合溶液,以不同的pH值以及鹽類濃度進行梯度冲提,得到逆流層析圖並每5mL收集一次,經過HPLC、MS分析各收集管中的蛋白質含量,可得到蛋白質相對濃度85 %的細胞色素C蛋白質溶液,有效回收率約為64 %,在製備蛋白質方面提供一新純化研究方向。zh_TW
